Specifically, the restriction sites were mapped as follows: (i) lambda DNA was cut using the restriction enzyme Handbill to form fragments of known base pair lengths which were separated by agrees gel electrophoresis; (ii) abrupt was digested in seven different ways using the combinations of restriction enzymes discussed above and the fragments from such digests were separated in the same electrophoresis; (iii) using the data from the lambda DNA fragments, a regression was run to determine the relationship between the log of the number of base pairs in fragment and the distance traveled towards the anode during he electrophoresis; (iv) the base pair length of the fragments from each digest was calculated using the relationship determined from the lambda DNA data; and (v) the length of the fragments produced by the different digests were analyzed to produce a map, as will be discussed below. It would take a thousand pages to describe the entire commercial and cultural aspects of the music industry, so I will talk about the hip-hop community. Some hip-hop is commonly referred to as Pop music, simply because it is popular with a wide variety of crowds. I intend to show you how the different types of crowds under the Pop category interact with one another, especially at popular awards ceremonies, such as the recent MTV Video Music Awards 00. MTVs largest show and party of every year is their Video Music Awards, celebrated in 1999 on 9/9/99. In 2000, it was highlighted by performances by Eminem (aka Marshall Mathers), NSync, a side of Britney Spears which brought a chuckle from a few early Madonna haters, and others. In an interview with reporters before the show, 26 year-old white rapper Eminem stated It aint often you get so many people that I dont like into one room together. Eminems relationship with the music industry is a strange one you either love him or you hate him. He openly hates gays, women, and children. He is being petitioned to be arrested by a national gay rights activist group for his homophobic lyrics and attitude. So, you ask, why is he such a loved character if he is so shady? His most recent album, The Marshall Mathers LP, sold 1.7 million copies its first week the most for any single artist in history. Also notable for comment is that he achieved this at a time when illegal music piracy is so common ly practiced that the government cannot even begin to regulate it. This is another issue however. The recent media chase over Shawn Fanning, an innocent-looking 19 year-old college dropout, is amazing for one who has not been along for the ride the whole time. Shawn Fanning released Napster in 1999, it was one of the top 10 most quickly downloaded programs of 1999. Its ability to share music files, called mp3s, digitally over the Internet made it become absolutely essential for anyone between the ages of 12-24 to have. Gone were the days when you had to listen to the radio or buy a single now you could just go online, wait anywhere from two to thirty minutes, and have the song that you wanted, for play on your own computer or to burn to a blank CD. Now it becomes just one of the many media-hyped incidents in the music industry. The pop music industry is a complicated web of underground culture. Because America is such a diverse nation, the music industry reflects the diversity of the country. Germany's Sex and Gender Roles What is it that makes up our identities? I mean us, not only Americans but other cultures as well, the entire world. When tracing back my family tree I learned that I am of half-German descent with a quarter French and another quarter Irish in me. I also think it is safe to say that a majority of the population reading this is of European lineage. I don’t say this to discriminate, only to be specific in the telling of my story. The story I am about to recite is about humans and about one of our basic needs for survival. Some will argue, saying that sex is not at all a basic need but let’s face it, without sex you wouldn’t be reading this intriguing article right now, and the world would be less fulfilled without the human reproduction process. Now that I have probably scared off the more ethical readers with my dirty talk I shall proceed. My story deals with the sex and gender roles that occur in the country of Germany. Why did I choose to study these roles in Germany? Well, it seemed like a good place to start, and being that I am 50% German maybe I can relate to some of these roles. We’ll see. When one chooses to study the sex and gender roles of a country it is quite easy for me to relate to the male population of the region. I believe that all heterosexual males desire the same common denominator from a relationship with another women. Yes, companionship is part of the desire but I have to be honest here, sex plays a major role in finding the right woman. So that was easy, I think most men in Germany agree with those two easy-to-read sentences stated above. Now here comes the difficult part, trying to figure out what women desire in a relationship. Being... ...n feel like she is in good hands. Can men who do housework protect their wives just as good? Maybe, but the West German women subconsciously saw these men as less â€Å"macho† than their eastern countrymen. So, did a cement wall allow one side of men to be more housewife like and the other side to be more manly? I believe it was not only the wall but also the government placed over the people. The easterners were frozen in a time where the men dominated and when that wall came down so did the western woman’s view of what a male should be. Their primal instincts drove them towards those dominating â€Å"macho† men of the East, and suddenly the fad is to acquire a man who will provide the maximum amount of protection, even if these men expected to be waited on by their newly conquered women.

Hypertonic refers to a solution with higher osmotic pressure than another solution. In other words, a hypertonic solution is one in which there is a greater concentration or number of solute particles outside a membrane than there are inside it. The red blood cells undergo crenation, which means they shrink and shrivel as water leaves the cells until the concentration of solutes is the same both inside and outside the red blood cells. Uses of Hypertonic Solutions Manipulating the tonicity of a solution has practical applications. For example, reverse osmosis may be used to purify solutions and desalinate seawater. Hypertonic solutions help to preserve food. For example, packing food in salt or pickling it in a hypertonic solution of sugar or salt creates a hypertonic environment that either kills microbes or at least limits their ability to reproduce. Hypertonic solutions also dehydrate food and other substances, as water leaves cells or passes through a membrane to try to establish equilibrium. Why Students Get Confused The terms hypertonic and hypotonic often confuse students because they neglect to account for the frame of reference. For example, if you place a cell in a salt solution, the salt solution is more hypertonic (more concentrated) than the cell plasma. But, if you view the situation from the inside of the cell, you could consider the plasma to be hypotonic with respect to the saltwater. Also, sometimes there are multiple types of solutes to consider. If you have a semipermeable membrane with 2 moles of Na ions and 2 moles of Cl- ions on one side and 2 moles of K ions and 2 moles of Cl- ions on the other side, determining tonicity can be confusing. Each side of the partition is isotonic with respect to the other if you consider there are 4 moles of ions on each side. However, the side with sodium ions is hypertonic with respect to that type of ions (another side is hypotonic for sodium ions). The side with the potassium ions is hypertonic with respect to potassium (and the sodium chloride solution is hypotonic with respect to potassium). Got it? Movement of Water in Hypertonic Solutions Water moves across a semipermeable membrane. Remember, water moves to equalize the concentration of solute particles. If the solutions on either side of the membrane are isotonic, water moves freely back and forth. Water moves from the hypotonic (less concentrated) side of a membrane to the hypertonic (less concentrated) side. The direction of the flow continues until the solutions are isotonic.
